Production
Genipin is extracted from the fruits of Gardenia jasminoides Ellis. Genipin is an excellent natural biological cross-linking agent that can be cross-linked with protein, collagen, gelatin and chitosan to make biomaterials. Genipin can also be used as a fingerprint collection reagent and a cross-linking agent for preparing immobilized enzymes.
Genipin is an aglycone derived from an iridoid glycoside called Geniposide present in fruit of Gardenia jasminoides Ellis.Genipin
is an excellent natural cross-linker for protein, collagen, gelatin, and chitosan cross-linking.It has much less toxic than glutaraldehyde and many other commonly used synthetic cross -linking regents.It has been used as a Chinese traditional medicine to relieve the symptoms of type 2 diabetes.It is also used for pharmaceutical purposes, such as choleretic action for liver diseases
control.
Genipin, as one of the popular approaches in controlling drug delivery from the polymeric carriers, is suitably achieved by the inclusion of crosslinking agents into the formulations at different concentrations. Nevertheless, addition of the chemical crosslinkers such as glutaraldehyde, formaldehyde etc, used in the drug delivery systems causes very serious cytotoxic reactions.
These chemical crosslinking agents did not offer any significant advantageous effects when compared to the natural crosslinking agents for instance genipin, which is quite less toxic, biocompatible and offers very stable crosslinked products.
Geniposide, the main active component of Gardenia jasminoides, belongs to the class of compounds called the iridoids, which can be hydrolyzed to genipin by β-glucosidase. Geniposide and genipin have been demonstrated to possess a number of pharmacological activities.

Increasing evidence also suggests that geniposide and genipin can improve the symptoms related to the progression of AD. For example, they have been reported to be associated with significant increases in learning ability, memory-related functions, and enhancement of cognitive function in animal models of AD. The addition of geniposide and genipin to neuronal cultures has been reported to antagonize cytotoxicity normally induced by Aβ exposure, oxidative stress, endoplasmic reticulum stress, and inflammatory reactions. These two compounds are reported to induce neurite outgrowth in neuronal cultures. This review will discuss the current state of the emerging research and discuss the potential development, use, and utility of geniposide and genipin as natural neuroprotectants for AD.
Application
Genipin can be used as a cross-linking agent for immobilized enzymes, as a regulating agent for drug delivery, as the
raw material for gardenia blue pigment preparation, and as the intermediate for alkaloid syntheses.
